However, the precise identity of SC-β cells in relationship to primary fetal and adult β-cells remains unclear. Here, we used single-cell sequencing datasets to characterize the transcriptional identity of islets from in vitro differentiation, fetal islets, and adult islets. Our analysis revealed that SC-β cells share a core β-cell transcriptional identity with human adult and fetal β-cells, however SC-β cells possess a unique transcriptional profile characterized by the persistent expression and activation of progenitor and neural-biased gene networks. These networks are present in SC-β cells, irrespective of the derivation protocol used. Notably, fetal β-cells also exhibit this neural signature at the transcriptional level. Our findings offer insights into the transcriptional identity of SC-β cells and underscore the need for further investigation of the role of neural transcriptional networks in their development. 000103656 536__ $$oNational Institute of Diabetes and Digestive and Kidney Diseases$$qhttps://ror.org/00adh9b73$$rROR$$cR01DK127497 000103656 536__ $$oNational Institute of Diabetes and Digestive and Kidney Diseases$$qhttps://ror.org/00adh9b73$$rROR$$cR01DK114233 000103656 536__ $$oJuvenile Diabetes Research Foundation$$qhttps://ror.org/0332bbh94$$rROR$$c3-SRA-2023-1295-S-B 000103656 536__ $$oHuman Islet Research Network$$cCatalyst Award 000103656 536__ $$oEdward Mallinckrodt Jr. Foundation$$qhttps://ror.org/035yf7a66$$rROR 000103656 536__ $$oWashington University in St. Louis$$qhttps://ror.org/01yc7t268$$rROR 000103656 536__ $$oNational Institute of Diabetes and Digestive and Kidney Diseases$$qhttps://ror.org/00adh9b73$$rROR$$cT32DK007120 000103656 540__ $$aCreative Commons Attribution (CC BY) 4.0 International$$uhttps://creativecommons.org/licenses/by/4.0/ 000103656 650__ $$aBiological sciences 000103656 650__ $$aBasic medicine 000103656 650__ $$aHealth sciences 000103656 650__ $$aHealth biotechnology 000103656 6531_ $$aSC-beta 000103656 6531_ $$a scRNA-seq 000103656 6531_ $$aTranscriptome 000103656 6531_ $$aGene networks 000103656 6531_ $$aDiabetes 000103656 6531_ $$aImmaturity 000103656 655__ $$aCode 000103656 7001_ $$aSchmidt, Mason$$1https://orcid.org/0000-0002-1303-9911$$uWashington University in St. Louis$$4https://ror.org/01yc7t268$$5ROR 000103656 7001_ $$aIshahak, Matthew$$1https://orcid.org/0000-0002-7494-6048$$uWashington University in St. Louis$$4https://ror.org/01yc7t268$$5ROR 000103656 7001_ $$aAugsornworawat, Punn$$1https://orcid.org/0000-0001-8602-5433$$uWashington University in St. Louis$$4https://ror.org/01yc7t268$$5ROR 000103656 7001_ $$aMillman, Jeffrey$$1https://orcid.org/0000-0003-0426-3492$$uWashington University in St. Louis$$4https://ror.org/01yc7t268$$5ROR 000103656 8564_ $$ePublic$$01$$9c749e42f-c549-4a6f-8b75-56e60921a825$$s2244247982$$2d25730f13d53bf104afc8aac3cffe8b9$$uhttps://data.library.wustl.edu/record/103656/files/all_endocrine.rds$$ySeurat objects containing the integrated CHGA+ cell populations from all datasets and the subset of beta cells 000103656 8564_ $$ePublic$$01$$9cca388b5-c676-4790-aafd-f74aa0dfbc8b$$s762376750$$2c69ca113df23e4598af82af17ec1b9d8$$uhttps://data.library.wustl.edu/record/103656/files/all_beta.rds$$ySeurat objects containing the integrated CHGA+ cell populations from all datasets and the subset of beta cells 000103656 8564_ $$ePublic$$01$$9f58f5416-e3c9-4b58-b29f-a4e33eabb28f$$s9944$$23f752fc1c8887c408d8f199b418f52e2$$uhttps://data.library.wustl.edu/record/103656/files/readme_doi1079366rxs103656_ishahak.txt$$yREADME 000103656 909CO $$ooai:data.library.wustl.edu:103656$$pdataset 000103656 980__ $$aWashU Researcher Data
